چکیده
To explore the involved mechanisms and possible treatments of ambient PM2.5 exposure-induced lung inflammation, this work studied activity luteolin, a natural flavonoid which widely presents in many plant species, murine alveolar macrophage MH S cells exposed to PM2.5. Results showed induced an inflammatory response, as evidenced by significantly increased TNF-α, IL-6, MCP-1 Rantes levels. iNOS, COX-2, NF-κB protein expressions MH-S cells. Moreover, luteolin pre-treatment reduced JAK2 STAT1 but not STAT3 PM2.5-stimulated Performing inhibitor AG490 further TNF-α IL-6 productions well expressions. In addition, although exposure could elevate HO-1 expression basically, administration enhanced additionally. Collectively, these results revealed that inhibits inflammation through suppressing JAK2/STAT1/NF-κB pathway enhancing PM2.5-challenged

Background: All cellular events depend upon the DNA synthesis and gene expression involving complex interplay between ligands such as interleukins and interferons, with various cell membrane receptors. These ligand-receptors interactions transmit signals within the cell via numerous signal transduction pathways to affect gene expression. Janus kinase/signal transducer and activator of transcrip...
متن کاملAllylpyrocatechol Attenuates Collagen-Induced Arthritis via Attenuation of Oxidative Stress Secondary to Modulation of the MAPK, JAK/STAT, and Nrf2/HO-1 Pathways.
Rheumatoid arthritis (RA), an inflammatory autoimmune disorder, is characterized by synovial hyperplasia and bony destruction. The pathogenesis of RA includes redox dysregulation, concomitant with increased levels of proinflammatory mediators. As the ability of allylpyrocatechol (APC), a phytoconstituent of Piper betle leaves, to alleviate oxidative stress has been demonstrated in patients with...
متن کاملProlactin-induced production of cytokines in macrophages in vitro involves JAK/STAT and JNK MAPK pathways.
Macrophages play a crucial role in host immunosurveillance against pathogens and malignancies. The enhanced productions of pro-inflammatory cytokines are central to the regulatory role of macrophages and induction of robust immune response. The excessive inflammatory response of macrophages can result into pathological conditions in host. We have previously reported that prolactin (PRL) induces...
متن کاملInfection-induced IL-10 and JAK-STAT
Generation of effective immune responses against pathogenic microbes depends on a fine balance between pro- and anti-inflammatory responses. Interleukin-10 (IL-10) is essential in regulating this balance and has garnered renewed interest recently as a modulator of the response to infection at the JAK-STAT signaling axis of host responses. Here, we examine how IL-10 functions as the "master regu...
متن کاملAnti-inflammatory effect of pristimerin on TNFα-induced inflammatory responses in murine macrophages
Purpose: To investigate the anti-inflammatory effect of pristimerin using RAW 264.7 macrophage cells. Methods: The effect of pristimerin on the RAW 264.7 cells proliferation was determined by CCK8 test. The expression of cyclin D1 and cyclin E1 in RAW 264.7 cells treated with TNF-α and pristimerin was measured by qRT-PCR. The regulation of cytokine IL-1β, IL-6, IL-8, and PGE2 in RAW 264.7 cells...
